Championship Burton made nine changes and were 1-0 down when Rashford dinked the ball over keeper Connor Ripley.
England forward Rashford made it 2-0 with a 20-yard finish before Jesse Lingard's deflected shot found the net.
Anthony Martial added the fourth from a Rashford assist, Lloyd Dyer scoring a stoppage-time consolation.
Elsewhere, it was a night devoid of shocks Chelsea put five past Championship Nottingham Forest, Everton eased past Sunderland and Arsenal beat Doncaster Rovers 1-0.
The closest to a shock came at the Hawthorns as West Brom scored a 72nd-minute equaliser against Manchester City before losing to Leroy Sane's second of the night.
